Physicochemical properties: white to light yellow crystal, red after exposure. It is soluble in hot water and hot ethanol with a solubility of about 1mg / ml. It is also soluble in alkali hydroxide, borax solution and glycerol, and slightly soluble in cold water and ether. When the solution is stored, the color becomes dark and the melting point is 100-120 ℃ (140 ℃). Irritating. Hematoxylin is one of the most commonly used dyes, which is a pigment extracted from the dried branches of the South American hematoxylin (tropical leguminous plant) with ether. Hematoxylin cannot be directly stained. It must be exposed to ventilated places to make it become oxidized hematoxylin (also known as hematoxylin). This is called "maturation". The "maturation" process of hematoxylin takes a long time, and the longer the time after configuration, the stronger the dyeing power. The dyed material must be treated with metal salt as a medium before it has coloring power. Therefore, mordant should be used when preparing hematoxylin dye. The commonly used mordants are aluminum sulfate, potassium alum and iron alum. Hematoxylin is an excellent material for staining nuclei. It can differentiate different structures in cells into different colors. The color of tissues stained during differentiation varies according to the treatment. After differentiation with acidic solution (such as hydrochloric acid alcohol), it turns red, and after washing, it still turns blue. After differentiation with alkaline solution (such as ammonia), it turns blue, and after washing, it turns blue black. It turns red when exposed to light. In aqueous solution, especially in alkali solution, it is easily oxidized by air to red brown oxidized hematoxylin.
