Purpose: biochemical research. Folic acid agonists. The synthesis of DNA is blocked by blocking the tetrahydrofolate cofactor necessary for the synthesis of thymidine. Methotrexate is transported into cells by folate transporters. In cells, it is converted into a high molecular weight polyglutamate metabolite by folylpolyglutamate synthase, and then bound to dihydrofolate reductase, thus inhibiting its activity. Methotrexate polyglutamic acid γ- Glutamyl hydrolase（ γ- Glutamyl hydrolasen) is degraded in cells.
