Physicochemical properties: white to almost white yellow powder, extracted by Aspergillus niger. Molecular weight: 150kda (about 80kDa in SDS-PAGE, isoelectric point 4.6). An oxidoreductase that oxidizes glucose to gluconic acid. The enzyme requires flavin adenine dinucleotide (FAD) as coenzyme, and each molecule contains two fads. It is easily soluble in water and completely insoluble in organic solvents such as ether, chloroform, butanol, pyridine, glycerol and ethylene glycol. It can be precipitated by 50% acetone and 66% methanol. The chemical substances EDTA, KCN and NaF did not affect the enzyme activity, but the enzyme activity decreased due to the influence of HgCl (mercuric chloride), AgCl (silver chloride), phenylhydrazine, p-mercuric chloride benzoic acid, etc
Purpose: biochemical research. The enzyme pair β- D-glucose showed high specificity. Desugation used in egg processing in food industry to prevent browning. It is used for deoxidation in beverages, fruit juices, canned and dried foods to prolong the storage period. It can also be used with catalase for food antisepsis and grape enzyme analysis. It is used as urine sugar and blood glucose reagent in medicine.
