Lysostaphin;Glycyl-glycine Endopeptidase CAS 9011-93-2
Item No.: E0028-1
Store at: 2~8°C
English name:Lysostaphin;Glycyl-glycine Endopeptidase
Other name: Lysostaphin
Grade:BR
Molecular weight: About 27kDa
Specific activity:≥1200u/mg for protein
Vitality definition:One unit will reduce the turbidity (A620) of a suspension of Staphylococcus aureus cells from 0.250 to 0.125 in 10 min at pH 7.5 at 37°C in a 6.0 ml reaction mixture
Determination of enzyme activity: By turbidimetry
Lysed Staphylococcus aureus: 4 ~ 32u / ml
Staphylococcus epidermidis: 8 ~ 64u / ml
Solubility: soluble in 50mmol / L tris HCl
Solution storage method:PH7.5 enzyme solution (pH5 ~ 8) is placed at 4 ℃ and basically stable within 2-3 days.
Physicochemical properties: White or light yellow powder, a zinc containing metalloproteinase, composed of 246 amino acids. The nucleic acid expressing the gene consists of 1486 nucleotides. The isoelectric point was 10.5-11.0, and the optimum pH range of enzyme activity was 10 ~ 11. Hydroxymercury benzoic acid (PCMB), benzenesulfonyl chloride (PMSF) and dithiothreitol (DTT) had no significant effect on the enzyme activity under the condition of 1mmol / L, and Zn ions showed some activation effect; Ba ion, Mn ion, Ca ion, Mg ion and EDTA showed a certain degree of inhibition on lysostaphin at the condition of 1mmol / L.
Purpose: Biochemical research. Bacteria that specifically degrade glycine peptide bond structure in cell wall such as Staphylococcus aureus can achieve the purpose of sterilization by cracking its cell wall.
