Application: the recombinant fusion protein containing glutathione S-transferase at the carboxy end or the protein dependent on S-transferase or glutathione was purified by affinity chromatography.
Purpose: pGEX fusion protein expressed in combination with glutathione S- transferase can be purified by glutathione agarose gel packing.PGEX is a kind of glutathione( γ- Glutathione (cysteinyl glycine) is used as a substrate to inactivate toxic small molecules of enzymes by forming mercaptan uric acid.Because the affinity of GST to substrate is sub mmol level, glutathione agarose gel affinity chromatography is effective in the purification of GST fusion protein. The binding GST fusion protein can be eluted with a buffer containing free glutathione.
Storage: sealed at 4-8 ℃. The used filler shall be thoroughly washed with pure water, and finally stored in 20% ethanol at 4 ℃.
matters needing attention:
(1) Before loading the sample, the sample must be filtered through the membrane and the pigment must be removed, otherwise the impurities and pigment will be adsorbed on the filler and affect the normal use of the filler. All buffers need to be filtered with a 0.45um filter.
(2) In the process of use, avoid using strong acids and bases with high concentration, and the concentration of acids and bases shall be less than 0.1 mol. Alkali slows the flow rate.
(3) Different samples have different adsorption and elution methods, which can be carried out according to the relevant literature.
