Physicochemical properties : liquid. The mutagenicity of cyanine dyes is significantly lower than that of EB, and it is safe to use. When combined with nucleic acid, it shows green fluorescence (the maximum absorption is 470nm), and the sensitivity of detecting nucleic acid is higher than that of EB. It can also be observed with ultraviolet lamp. When the nucleic acid is treated by conventional ethanol precipitation method, the dye can be completely removed from the double stranded nucleic acid.
Purpose: biochemical research. SYBR Green 1 is a highly sensitive DNA fluorescent dye suitable for various electrophoretic analysis. It is simple to operate, without decolorization or rinsing, and can detect at least 20pg DNA, which is 25-100 times higher than that of EB staining. The fluorescence signal of SYBR Green 1 binding to dsDNA will be enhanced 800-1000 times. The gel sample stained with SYBR Green 1 has strong fluorescence signal and low background signal. It can be applied to a variety of electrophoresis analysis. SYBR Green 1 is applicable to a variety of gel electrophoresis methods: agarose gel, polyacrylamide gel electrophoresis, pulsed electric field gel electrophoresis and capillary electrophoresis. SYBR Green 1 has a very high affinity with double stranded DNA, so it can be used for pre electrophoresis staining, and has no inhibitory effect on enzymes commonly used in Molecular Biology (such as Taq enzyme, reverse transcriptase, endonuclease, T4 ligase, etc.). In addition, compared with EB, SYBR Green 1 has a greatly reduced mutagenic ability.
